Good day! I am trying to find a way to measure hesperidin content of the peels of my fruit sample (Citrus reticulata), other than HPLC. I plan to ferment my peels with food-derived mixed cultures (like fermented tofu and kimchi), and monitor and measure the hesperidin content each fermentation days. We only have one HPLC instrument in our laboratory, and a lot of us are going to use said instrument. Considering the operation of HPLC can be time consuming, measuring my sample's hesperidin content using HPLC could be impractical, which is why I am trying to find other ways to measure.
There are several alternative methods for measuring hesperidin content in fruit samples other than using HPLC. Here are a few options you can consider:
1. Spectrophotometric Method: This method involves extracting hesperidin from the fruit sample and then measuring its absorbance at a specific wavelength using a spectrophotometer. You can follow a well-established protocol or use a commercial hesperidin assay kit.
2. Titration Method: This method involves the use of a specific titrant that reacts with hesperidin to produce a color or turbidity change. The endpoint of the reaction is determined by observing the color change or other visual cues. This method requires the use of a standard curve to quantify the hesperidin content in the fruit sample.
3. Fluorometric Method: This method utilizes the fluorescence properties of hesperidin. By exciting hesperidin with a specific wavelength of light, it emits fluorescence at a different wavelength, and the fluorescence intensity can be measured using a fluorometer. Calibration with known concentrations of hesperidin is required to determine the hesperidin content in the fruit sample.
4. Liquid Chromatography-Mass Spectrometry (LC-MS): Although this technique is somewhat similar to HPLC, it provides the advantage of simultaneously identifying and quantifying multiple compounds, including hesperidin.
