I am a rookie in cell biology and I am performing an overexpression of my target protein in the HeLa cell. The expression vector I am using is pCI-neo. I have observed some puncta subcellular structures, but I am not sure whether they are organelles that the endogenous protein located, or if it is just the protein that aggregates when overexpressed. I have tried to determine the puncta structure by labeling the marker of its destined organelle(stained blue) and some cells fit well, but others are just partially co-localized. Will protein aggregates also look like puncta structures under the confocal microscope? Have a look at my attachment. Do you have any suggestions about the aggregates?
