I am trying to replicate results I see in-vitro, with cells cultured with 1,25 dihydroxyvitamin-D (1,25 VitD) but by getting skin biopsies (from healthy donors of surgical procedures) to produce the VitD.

I have tried sitting an 8mm biopsy punch in a transwell (with the bottom of the skin emerged in media in the top of the transwell) and shining UVB light on the skin, but this does not yield 1,25 VitD. 

Is it possible that the skin retains the active Vitamin-D and absorbs it in the keratinocytes etc?
