If you're using qiagen kit , you can increase the incubation time before elution from the column,, try it, iam doing this to increase the yeild of DNA from paraffin tissue

Yes. You can extract from buffy coat to increase your DNA yeilds. We are doing this in our lab with Qiagen kits and it is working great.
