I have been using a t test to do it, considering homoscedastic and two-tailed t test (acceptable level of more than 70%), but I don't know if it is appropriate...

Also, I don't know how much concentrations should I use to do this comparison. I mean, I know that in common validation method I should use only 3 concentrations (5x each), but for this type method I don't know if is enough ou I should use all the 5 linearity concentrations (5x each).

For the matrix effect also 3 concentrations are enough?

I will be very grateful if someone could help with those questions...
