I could not see the protein bands very clearly when painting coomassie Blue.
The factors involved are the coomassie blue dye concentration, period of exposure, period of destaining and most importantly the amount of your protein and weather it is still sound or denatured. My suggestion run more protein.
Dear Savari
Thank you for your guidance.
The concentration of your loaded protein goes a long a way in determining the intensity of your bands
Hi Thomas
The protein concentration problem is not high because of the high concentration of protein loaded.
I do not know what is the problem !!
In fact, with coomassie Blue coloring in comparison the silver nitrate shows a lower concentration of protein.
When I work with this method, all proteins are not completely transferred from the gel to the membrane (PVDF), and after staining, there is still some protein remaining on the gel. I'm looking for...

I got these smeared bands quite often lately. We typically run the gel at 140V with a 10-12% gel and do a wet transfer at 220 mA for 1.5 hr in cold room. We also noticed some dirty spots/dots (see...
