Have you checked the phenotype of the cell expressing the entire fusion and/or the phenotype of the cell expressing  the proteinA with tag or proteinA alone are different from the one you have with the truncated fusion?

Thank you for the links. I have checked these sources, but couldn't find any motif or signal. Maybe these extra amino acids have an affect on the stability of Protein A which needs further investigation.

I have checked the phenotypes of the cells expressing Protein A, Protein A+Tag or Protein A+Tag(partially)+ELCRRLLL.

Only the last one results in an interesting phenotype.

Is it a complementation assay for proteinA biological activity or just an expression experiment? Are the three proteins expressed efficiently?
