i prepare protein nanoparticles. in one of my synthesis i use trifle amount of crosslinker and the nanoparticles stick in the bottom of plate. how can i understand that nanoparticles become crosslinked or not? through which analysis i can achieve this fact?
Dear Elham Saleh, if you know a good solvent for this protein, do solubility test and see if it is still soluble. If you have only swelling then you may say that there is crosslinking. My Regards

It is just a qualitative test with an arbitrary amount of solvent, because you just want to see if crosslinked or not. Of course the amount of the solvent should be much higher. If it is crosslinked and you want to quantify the degree of crosslinking then precise amounts are to be taken, to get the crosslink density via swelling test, which is widely discussed in this platform (just make search within RG in case you want to know more and how it is performed practically). Do solubility/swelling test. If soluble, they may not have cross-linked. Swelling shows they have actually cross-linked
