This a three DNA extraction methods from a single lymphocyte or blastomere and compared amplification efficiencies DNA extraction by :
(A) lysis in distilled water with freeze-thawing and boiling;
(B) two-step lysis involving potassium hydroxide and dithiothreitol;
and (C) the spanning protocol, using N-lauroylsarcosine

there are several ways, if you do one or two amplifications you can just do PCR with one cell in tube, If you are looking gor genome amplification - ther are Lyse-and-Go protocols
is there any protocol to purify PCR products without kit or column, I have small amount of PCR and I dont want to lose it, can I use phenol/chloroform and etoh precipitation for this purpose? thanks
