I have been trying to dilute Cisplatin in 0.9% PBS, but when ever I dilute it instead of completely getting diluted the tube resides some yellow colored minute powdered particles, they get suspended to the bottom even after vortexing.
I have tried using a water bath sonicator to dilute it, once sonicated for 5 min at 10X limit, the particles are not visible, but if the tube is left still at 4 Degree after sonication for about 1 hour or more the particles come back to light.
Due to this I am perfectly not able to figure out if the drug is getting diluted or not.
Kindly share ur experiences using cisplatin (powder form)
It is possible that the PO4-3 and /or Cl- in PBS is displacing the NH3 from the Pt core in cisplatin. That said -can you try a different buffer composition - perhaps HEPES or Tris at the same pH/ionic strength?
Hi Gautam. Try 0.45% NaCl which is the best solvent of cisplatin as far as I know (1 mg/ml). Also you can try 0.9% NaCl which id clinically approved and the solutions will be stable for more than 2 weeks at 4 degree. NB:Never try diluting in DMSO as it inactivates the drugs potency. PBS is not a good solvent for cisplatin but you can make lower concentrations without precipitation (
