I read that i should first differentiate SH-SY5Y with RA and then transfect them. How long should i wait before i transfect this cells?

Thanks for your answer. I have just one more question : how long did you wait before transfection ? In the litterature, i read that SH-SY5Y will be differentiated after 5 to 7 days of RA treatment. Should i wait this long to transfect my cells ?

For transfecting differentitated SH-SY5Y I recommend NeuroMag as you can see it has been very good at transfecting all kind of primary neurons and neuronal cell lines such as yours

I also work with SH-SY5Y. We differentiate them: 4 days TPA + 4 days RA.
I was transfecting the cells after 7 days (keeping them on RA) .
The efficiency of transfection lipofectamine 2000 dropped dramatically after I started differentiation. So I switched to nucleofection (Amaxa, Kit V) and it works just fine. For me is worked just as they claim in the protocol:

I have a question I am trying to transfect my SH-SY5Y after differentiation and after I transfect I keep them in 1% FBS with the RA. They died like crazy. Do you put them back in 10% FBS? Can you explain me what do you do?
