Extract Chondroitin Sulfate from the formulation matrix. It is soluble in water, but not in alcohol. Dissolve the matrix in methanol with just enough water to get all in solution. Than add small amount of water to reprecipitate the gel/cream and centrifuge the mixture to get the dissolved Chondroitin Sulfate for HPLC analysis. Make sure you measure all the volumes to determine the concentration accurately.

You can try ethanol or IPA instead of methanol. Also use different ratios of alcohol and water to get full recovery, not only 65%. Sonicate before reprecipitating the gel/cream.
