Constitutive enzymes are expressed all the time

Measure the expression and activity of the enzyme under the "normal" condition and the "adaptive" condition. An increase under the adaptive condition would suffice to describe the enzyme as adaptive. However, if your are measuring an increase in the activity of the enzyme, it is important to make sure that the adaptive condition is not just the presence of a higher concentration of the substrate of the enzyme with no change in expression of the gene for the enzyme.

Normally any organism produces two types of enzymes either constitutive or inducible based on the metabolic requirements of an enzyme..... All the enzymes which are involved in those processes which are essentially required for various metabolic activities are constitutive. Irrespective of whether the substrate is fed or not, they are expressed in the organism and constitutively being produced. There are certain enzymes which are produced/ whose gene expression happens only when the substrate is available to them. Such enzymes are referred to as inducible enzyme....  Adaptive enzymes I have no clue about..

I can measure activity under adaptive condition, but how can I measure enzyme activity under normal condition?

I think there isn't any different between inducible and adaptive enzyme. Adaptive is another name of inducible

If the enzyme is present in the constitutive growth condition and there is a selective assay for it, then you can measure it's activity in the medium. As a baseline, you can either use medium that has not been used to grow cells, or medium that has been used to grow cells but from which the enzyme has been removed by ultrafiltration.
