Islets will sometimes become attached via fibroblasts, but not always. You will need to coat the plates with a matrix to improve attachment. I have never done this, but you should be able to find references in the literature. Why do you want them attached? If you are doing secretion studies, you could place islets in mesh bottomed tubes. I used to make such tubes from nylon mesh and polypropylene 12 x 75 test tubes. You can cut tubes into sections and briefly heat one end and then press onto a piece of mesh. The melted plastic "glues" the mesh in place.

I am trying this experiment to measure the insulin level.

I found some islets attached to the plate 2 days after isolation . But I want all islets to be attached. Next time if I find less number of islets again then I will try to coat the plate with matrix or can try for mesh bottomed tubes. I may ask you again for further information.

I am again facing trouble with pancreatic islets isolation.

This this I found most of the islets are broken and only few islets clamped on mesh after centrifugation. I did hand shaking few times during collagenase digestion to make homogeneous suspension.

I don' know the broken islets are due to hand shaking or centrifugal force.

Hello Sharmin Akter , if your islets are broken you might use too long a digestion or shaking for too long. 12-15 (max) minutes of collagenase (0.95 mg/ml) digestion at 37 degrees, followed by moderate shaking for 1 min max, should be more than enough to isolate the islets.

Sharmin Akter, I also forgot that I had similar adhesion issues that I solved using Cell-tak. It is very strong and it worked even after only 2 hours from plating.
