For how long can human non cancerous pancreatic cells survive at room temperature under a microscope? I would really appreciate it if anyone with experience can tell me if their cells survived in this condition. The microscope was on and the cells stayed for about 3 hours.
It is not impossible to keep it at room temperature for 3 hours, but there is damage to the cells. The first thing to do is to proceed with the experiment, keeping in mind that the results may be a bit artificial.
Assuming that the cells are placed in room temperature conditions, the CO2 concentration will probably be the same as in the atmosphere. In that case, the pH of the medium would change significantly and the effects would be quite severe. In that case, adjusting the pH with HEPES (at the desired temperature) will avoid the effect a little.
When I used a sea horse flux analyzer in the past I could control the temperature but not the CO2. At that time I used medium with HEPES depending on the case.

we ship cancer cells lines cultured in flasks @RT on a regular basis. These will be @RT for at least 24-48hrs. No problems. If it's much longer or colder, they'll suffer. I wouldn't worry much about 3hrs @RT. It won't hurt to run one of your standard assays though to see whether they still behave as they should.

I agree with Thomas Mühlenberg. The only problem is the pH of your media with prolonged exposure to air, as when I would ship cells, I would fill the flasks completely with medium. The medium should be changed to fresh and cells returned to incubator and accessed the next day as some cell lines are more tolerant than others of pH changes. Best of luck!
Although you can mitigate pH changes using HEPES or other appropriate buffer, I would be more worried about the duration of the light exposure heating the sample above 37 deg C. If the light is turned off between users, there should not be much of a problem, but if you have student after student looking at the constantly illuminated sample, there could be a problem.
