well as you read the question, I want to coat PLGA on the polystyrene plate for antibody detect with ELISA assay. Is it possible to use usually way for peptid coating? because PLGA is not soluble in non organic solvent like bicarbonate buffer and i think it make difference for coat it. Can DMSO make interference for coating? so what is your comment? is there any solution for coating PLGA?
I would try to prepare a DMSO stock of it and dilute into water, PBS or whatever you like. If this should not work, try an organic solvent which may be easily evaporated (not DMSO or DMF, but does not dissolve your microplates - halocarbons and acetone etc might be a bad choice). If everything fails, couple your antigen to a carrier protein like BSA and use this for coating.
Breaking down the polymer into shorter pieces by ester hydrolysis (or homemade synthesis) might be another approach. Antibodies against PLGA should recognize shorter fragments.
