As far as I know, IDO1 is expressed inside the cells (ie not a secreted protein). I am afraid that serum/plasma is not a suitable sample type or the level of IDO1 may be too low to be detected in serum/plasma.
Yet i am wondering if there is any extraction method available for measuring the expression of IDO1 in serum? Currently I am using an ELISA kit to measure the expression of IDO1 in cells. Any pre-Elisa steps for Serum to check the IDO1?
Note: I don't want to check serum IDO activity for example; Kynurenine/Tryptophan ratio.
You can also confirm it by western blot experiment after preparing cell lysate.

Adnan Khan Although, I did not work on IDO1 expression. However I did work on expression of other metabolites. If you already have antibodies to detect IDO1 expression that you use for Elisa as well.
