A vector can accept an insert of 20 kb size and the recombinant vector can be replicated in E. coli. In order to make complete gene library of E. coli by using this vector, what is the minimum number of bacterial colonies required..?? How to calculate this ??

It depends on the nature of your DNA insert.
If its a variant library that you're looking to construct then the maximum possible of colonies but you'll be limited by your screening capacity thought.
If its only a unique DNA sequence then one colony is enough. But you'll need to verify correct insertion by Agarose electrophoresis before proceeding with any other work.
