Can I use Livak method to compare the expression of 1 gene of interest (X) between 2 species of bacteria (group A streptococci vs group B streptococci)?
Y gene in both 2 species was use as refference gene (housekeeping gene).
The formula is 2^-delta delta Ct, where delta delta Ct=( delta Ct of treated - delta Ct of control). Delta Ct treated= (Ct of target gene in treated group - Ct of GAPDH or, beta actin or, 18s gene in treated group) and Delta Ct control= (Ct of target gene in control group - Ct of GAPDH or, beta actin or, 18s gene in control group). You can see the paper of Livak.
Thank you for your answer Laurence Stuart Dawkins-Hall
I have 2 more questions that need your help.
1. Which one should I use for presenting relative gene expression between control and treated group dCt, ddCt, 2^(-ddCt), log2 of fold change?
2. I found that some paper used "fold change" and some others used "relative expression of mRNA" in thier figures. Is there any difference between them?
