I have taken 2 mg leaves and crush in 5 ml solution.Actually i have to take 2 mg leave sample and 2 ml solution.
So during calculation how can i calculate df in my formula. Smilarly if i add my dilution in extract then how can i calculate.Please guide

i am taken 0.2 g leaves and added 7 ml buffer solution.
According to the method i have to use 0.2 g leave with 2 ml buffer solution
.So how can i calculate dl value?

The dilution factor is more commonly used to compared a concentrated starting solution that one would dilute to make it less concentrated (i.e. adding 1 part concentrate to 4 parts solution would give a dilution factor of 5; this is written as a 1:5 dilution).
If I understand correctly, you've made a solution from your leaves that is too dilute for your method (~0.029g/ml compared to the required 0.1g/ml). To find out how dilute your sample is, I would compare these two concentrations by dividing the more concentrated solution by the concentration of the solution you made:
0.1/0.029 = 3.45 (dilution factor)
This means that the solution you made needs to be 3.45 times more concentrated (or conversely your solution is 3.45 times more diluted). I'm not sure if that method needs a specific concentration, mass or volume. If you just need a specific mass of leaves in your method than you can add more of your dilute sample where needed, so if the method asks for 1ml of leaf solution, you would add 3.45ml (1ml * dilution factor). This, however, could mess up your method very easily if specific concentrations of other reagents are needed. I would rather just make up a new solution at the correct concentration.
You could also try evaporating the buffer but that would also concentrate the salts contained in solution, which could effect your results so I would not recommend that. But it really depends on the method your are using and what you are testing for.
