I have used the substrate "ABTS" (Sigma) which needs a "wavelenght correction set at 650nm", as specified on the Data sheet. I think the plate reader (BioRad) is quite old and I haven´t been able to find this option in it. I don´t know whether it would be correct to calculate the subtraction with Excel. In any case I have tried that and apparently is fine for the absorbances, but the resulting concentrations doesn´t make sense.

Yes, you could read the plate at 650nm and at the other wavelength and then, to make manually the corrections in Excel, but you must do the corrections in both: your samples and the standard concentrations curve. If even so, resulting concentrations aren’t logical for you, then I advise you to do the test again.
