I want to remove non-templated nucleotides that are added to the 3' end of my cDNA by reverse transcriptases, creating blunt-ended RNA/DNA hybrids. Standard blunting protocols (T4 DNA pol./Klenow + dNTPs) seem to be chewing back too many positions, creating 5' overhangs. Does anyone have any experience with this?
