I want to attach a TMB dye to an antibody using a peptide based linker that can be cleaved with a peptidase, so how can I attach the dye to the N- and C-termini or to amino acid side chains ?
You can able to attach it, but before that you choose active site exactly where you are attaching TMB for your function of study, if you are going to attach in C-terminal means it give reverse effect no use, only you need to attach N-terminal. if you are attaching in amino acid side chain means it won't function.
before answering this question, need to know, whether your peptide active N-terminal was free of it have to bind some where, this thing you need to concern. after attaching dye whats next step, going to express? or bind! because after blocking N-terminal with dye means not possible to bind others on it.

so I want to attach this dye to an antibody using a peptide based linker it doesn't mater whether its attached in N- or C- terminal I just want it to be delivered with the antibody and then be released when I add the peptidase , nothing will be bound on it and it doesn't matter if it wont function as long its still attached to the antibody (once released I`ll need its function).
