Does anyone have suggestions on home-made non-denaturing lysis buffer/or cheap commercial source that can be used for fluorometric plate based assays?
Similar to the assays that Promega sells, I want to be able to add the lysis reagent along with a fluorometric substrate, incubate, and read the plate. The Promega version of the assay we want to use is quite expensive, but we can get the substrate relatively cheap. Any suggestions?

Here is one example of a non-denaturing lysis buffer:

20 mM Tris HCl pH 8
137 mM NaCl
1% Nonidet P-40 (NP-40)
2 mM EDTA
It may have some fluorescence at short UV wavelengths, similarly to tyrosine, but should not cause interference at visible wavelengths.
