I want to use the dental pulp tissue to do some work on stem cells. it is assumed that since they arise from neural crest cells, they might be useful for research into brain diseases research.
After extraction of tooth, you can use sharp excavators to swoop the pulp from tooth opening. Then you need to do repeated washings in PBS. Chop the tissue into minimum fine pieces and then do further washings with PBS (2-3X). After washing you can transfer the explant pieces to 6-well plate contining 1ml complete media or you can process the tissue for collagenase digestion. After collagenase digestion, take care to pass the cells through cell strainer and give a washing in media. Please see recent papers published by me which you can find in my profile.
The main thing to take care is proper washing of pulp tissue and explants with PBS and you won't face any contamination problem.
Before opening the tooth, you can keep the tooth in 1xPBS containing antibiotics and antifungal (double the dose) for 20 minutes and then follow the normal procedure for explant culture.
