The tests for any bacteria? general , The acid production from carohydrates was uesd nutrient broth without glucose and add 1% from any carohydrates found chlorophenol red indacter after 3-5 sdays . the charge red colore to yellow colore was poisitive
The aboves are possible by API test strips. If you need mannual test,
1. Acid from ~ : prepare medium (Peptone 10g, Beef extract 1g, NaCl 5g, Phenol red 0.018g with 0.9 liter DW, pH 7.4) and autoclaving it. -> filter sterilize 10X sugar solution and add previously prepared medium (final 0.5 (salicin) or 1% substrate) -> mix thoroughly. Or prepare in a whole and sterilize it at 115 Celsius degree for 15 min. (conc. phenol red solution could be prepared with 10% ethanol). You can add ~0.5% agar for semi-solid medium.
This is the formalized methods, you can change NaCl conc. etc. depend on your strain.

simply try 2% Peptone, 1% respective sugar in media thats all (for reffinose, double the concentration)..
to detect acid production you can use andrade's indicator so that on acid production media will chnge colour to pink. or can also try some indicator dye like bromothymol blue or bromocresol purple etc..
for detection of gas one can add durham's vial.
You have all or most of them at the same time in API20E AND API50CH strips (BioMérieux) !!
