I try to isolation of hepatocytes from mice but I didn't get viable cells. I tried almost 4 to 5 different protocols and Collagenase 1 and 4 but it wouldn't help. Please give me suggestion what are things I need to do to getting enough yield of viable cells.

Article Protocol for Primary Mouse Hepatocyte Isolation
As mentioned by the investigators, this protocol shows better reproducibility, shortened duration, reduced technical challenge, increased yield and higher viability. The type of collagenase used (i.e., Liberase) shows quicker digestion and better reproducibility compared to other collagenases.
As expected outcome, the typical hepatocyte yield is 30–50 x 10^6 viable cells per mouse and viability is approx. 95%.
