17.5 mg of collagenase in 50 ml of Ca-containing buffer (P2, freshly prepared). Before collagenase perfusion, it is preferable to perfuse with HEPES buffer (P1), that is free of calcium. Try to oxygenate the buffer and maintain a temperature of 37°C throughout the perfusion process.
