I'm working with ETH-2-Gold phage and VCSM13 helper phage, I'm using TG1 as my host. I spin down the bacteria 10,000rpm for 30mins, then add 2ml of 20% w/v PEG6000 in 2.5M NaCl to 10ml of supernatant and chill on ice for 1 or 2 hours. I again spin down at 10,000rpm for 30 mins and sometimes I get a plaque...it's very hit and miss. I sometimes increase the amount of PEG but that rarely works if I didn't get the pellet the first time.

Any help would be greatly appreciated :)

*Bonus question: I have both TG1 and XL-1 blue strains of e.coli but only use TG1 exclusively at the moment...is there a big difference or does it matter at all? Thanks.
