Hi,

From right left I assume 5' and 3'. If your protein coding gene is 5' or left promoter will be on left (5') of gene. If your TSS is on 3' (right) promoter will be on 3' (right) end.

For example we have two gene

Gene1 1000-1500 +

Gene2 2000-2500 -

For Gene1 promoter will be in between 1-1000 as + indicate "+" strand (5' to 3') and for Gene2 promoter will be 2500+ (say from 3000 to 2500) as "-" indicate 3' to 5'.
