Hi Sajib,

In my experience, it was very difficult to transfect such large plasmid into bacterial cell if using usual transfection reagent such as Superfec or Polyfec. Our previous succed to transfect was only for 3.2kb plasmid. Larger than that, I recommend you to use electroforation method that can be used up to 40kb, in our experiences.

For a plasmid of that size, I would be doing electroporation linearizing the plasmid. Hope this helps. Good luck.
