Have you had problems with the solubilization of 4-Methylumbelliferyl α-D-glucopyranoside?

Hi guys,

I want to perform an alpha-glucosidase activity assay, but I have had problems with the solubilization of its substrate 4-Methylumbelliferyl α-D-glucopyranoside.

I purchased 4-Methylumbelliferyl α-D-glucopyranoside from Sigma (M9766) and tried dissolve it at 1,5mM in 2mL (0,507mg/mL) in buffer phosphate-citrate 0.2M. I tried incubate the solution in water bath at 42 C, but I couldn't get a completely solubilizaded solution.

Any ideia about what is happening and how I can solve this problem?

You might want to try N,N-Dimethylformamide (DMF) or DMSO as the solvent instead. Sigma lists the solubility at 50 mg/mL in pyridine so I'm guessing the compound isn't particularly soluble in water.
