I am interested in doing an experiment on exosome research. I want to attach a fluorescent tag to exosomes or vesicles extracted from cells, and then put the exosomes or vesicles back into another cell (perhaps a cancer cell) and track where it goes. So I would need to use 2 fluorescents, one to attach to the exosome or vesicle and one to attach to the cancer cell.
Hi Chloe, you can label the exosomes with dyes such as PKH 26 (HY-D1451) or PKH 67 (HY-D1421) before adding them to recipient cells. After uptake, you can stain the cells with another fluorescent marker to distinguish cellular structures and track where the exosomes localize. For example, this study (PMID: 32495906) used DAPI to stain nuclei and Rhodamine–phalloidin to stain the cytoskeleton, then visualized the co-localization to determine exosome uptake.
