I am using the BB1 receptor antagonist PD168368 at 10uM as has been shown in the literature, but am having trouble adequately inhibiting activity through this receptor in the basolateral amygdala of SD rat pups.

I first attempted to preincubate the slices for 1 hr prior to patching and recording, but did not have success. In fact, preincubation seemed to augment the effect of 0.3uM NMB. Next I included PD168368 in the all solutions (external recording solution, external recording solution with kyneurnic acid, and in external+KA+0.3uM NMB). There still was a normalized increase in the frequency of sIPSC events.

Should I increase my incubation time or treament of slices prior to drug (NMB) treatment? Should I increase my concentration of PD168368, if so, to what concentration?
