I would like to try protein expression in HEK293S GnTI- cells but in suspension culture. Has anyone tried adapting adherent HEK293S GnTI- to suspension conditions?
I'm routinely using the standard Expi293 (you can find a presentation about it on my blog PRoteoCool ( https://proteocool.blogspot.com/ )
The system is quite expensive, especially the transfection reagent but they work very well (for some lucky proteins i obtain up to 500mg/liter yields in shake flask)
I'm never tried the Expi293 GnTi- cells but some former colleagues, that are currently using it, told me that work quite well and very similarly to the standard Expi293.
Of course if you would like to save money you can try to use it with alternative media or transfection agent but i'm not sure that you will obtain similar yields.
i performed some prelininary t FectroPro transfection agent (which is quite cheaper that expifectamine). It seems to work quite well but my data are still too preliminary to be conclusive.
