I am trying to run an LDH on fibroblastes cultured in media containing 10% serum. The literature from our lab states that past studies have shown that LDH will not work when run with media containing serum. The company which manufactures the assay has stated that the serum level must be 2% or less. At the moment, I am trying to determine whether I should consider attempting to grow the cells in serum reduced media or whether anyone has successfully run the LDH with 10% serum, or whether there is a similar assay that can be used with serum?
Hi Christiana. I've always used 1% FBS with HUVECs cells. I think you should try different % of serum and see if there's a difference. Good luck.

The behavior of LDHs will be affected by the dormation of the protein corona for sure. We have performed some studies on the change in the interfacial properties with the adsorption of albumin and there are significant changes. This will for sure affect the internalization capacity of LDHs. We are currently performing studies on the protein corona formation onto LDHs in serum and their effect in internalization capacity. However, the point is to what extent the results in cell cultures are significative if LDH interactions with serum are neglected.
