I've stained cryosections on non-coated positively charged slides with Alcian Blue with prior success but since the tissue was unstable for IHC, I sought to coat my slides with 0.5% gelatin, 0.05% chromium potassium sulfate (as per RnDsystems protocol). I usually stain sections in 1% Alcian Blue, 0.1N HCl overnight, but since using the gelatin-coated slides, none of the sections stained. I'm wondering on the positive charge of the chromium potassium sulfate may be interfering with the Alcian Blue dye? Or perhaps I need to pretreat my slides with something before the dye can stick? Thanks for any thoughts!
