My lab is currently using the manual column method for dual extraction of RNA and DNA from FFPE tissue (from the same sample) for NGS . This is due to the limited tissue available.
The Fisher Recoverall and Qiagen Allprep kits allow this, but they are manual column extraction methods
Does anyone know a reliable and compact automated extraction system which can do this (dual RNA and DNA extraction) ?
Dear Christina column based methodologies are not much efficient for RNA/DNA extractions from FFPE rolls both in terms of quality and quantity. Following kit is one of the best in FFPE case for both DNA and RNA and it can be used manually as well through automation. I have used this kit several times manually and through automation for FFPE sections without having any compromise in RNA / DNA yield and their quality. It is magnetic beads based technique with quite good and more often unimaginable results, when the sample is the notorious FFPE. Especially, the RNA extracted by this kit I have used further for downstream applications without any optimizations in my qPCR protocol, which I never observed in column based kits in the past.
