I've got a plasmid from SinoBiological- Casp-3 GFPspark cDNA clone. In the section of expression method it says- Constitutive, Stable/transient. I would like to know what ''stable'' is here? I want to create a stable cell line using this plasmid
Stable, reffers to the clone having been made and is expressing the gene of intrest from its genomic region from the transformation process. It must be keep under slective presssure like G418 to keep the insert.
Where as a transient would be having used a transfection reagent and mix it with your plasmid expression vector, and then add it to a monolayer of cells and have them express it for a short time before end point and assay.
You may want to look into Lipid based transfection reagents. Like Lipofectamine and use this to place your plasmid into a cell line, to make your own stable expressing cells.
