I have done RT -PCR for Cyclin D2, p21 and p27 in du145 , but did not get any band . in p21 , there was v. faint band but when I repeated the same , I did not get any band for p27. does these 3 genes show expression in du145 cells or not? and what is the status of methylation of these genes in DU145 cells and LNCaP cells.?

Are your primers working? Have you tried using them with another cell type? Is your RNA good? (high 260/280 ratio, no degradation) have you successfully amplified an endogenous control using your RNA?
