I have used 1% penicillin/streptomycin and 20% FBS to supplement the M199 media. Those organisms are slow-growing and later on affect the growth of the Leishmania parasites also. Usually they take more than one week to grow. Samples collected from different patients (lesion aspirates and slit skin samples) had the same issue. Also used fresh media and consumables.
Please let me know how to get rid of this.
Hello Rajamanthrilage. Is it a culture of promastigotes? If so, they may be fungal contamination. You could prepare a quick Giemsa staining of your sample and verify that those cells do not have kinetoplast. 5-fluorocytosine has been used to culture Leishmania and prevent fungal contamination during initial isolation or field conditions.
Agree with Marlene Jara. We found many like these when testing microcultures. Not entirely sure if they are bacteria or fungus but a Giemsa or Gram stain could be useful to figure it out.
If they don't move, probably are fungus.
If you mean small moving organisms, which appear to be bacteria, we have the same problem, but we are not sure what it is. They look like streptococcus, but are 5-10 times smaller than leishmania. I don't know if you are speaking about the same thing.
