We are using the Bio-basic SEC 60 column to further purification of protein fractions using HPLC analysis. Sample initially separated via membrane filtration and less than 1kDa fraction was used as a feed sample for the HPLC. The sample (200mg/ml concentration) was elucidated with 100% water. When continuous operation column pressure was increased gradually (around 500 psi to 1600 psi at 0.5ml/min constant flow) and recently column pressure is too high to work on flow rate 0.5 mL/min. Are there any suitable methods to clean the column (Bio-basic SEC 60)? I washed the column using 100% water at 0.5ml/min flow but it is not sufficient to clean column and the pressure still was too high. Are there any suggestions on how to restore the column?
