Also, is it regular MTT assay (formazan or MTT kit) will work properly in spheroid case?
Hi. You can definitely create a graph either by using GraphPad or sigma plot. Yet, to show the size, it is always advisable to capture the pictures using a microscope. This will help you in showing a better representation. Regular MTT will not work for spheres. You might probably need to use the spheroid kit to perform that.

To determine the number and area of your spheres, you can take micrographs at a wide-field objective (20x, for example). If you grow them in 24-well plates, you could take random catches from 8-10 fields. Using ImageJ, you could delimit each spheroid and determine its area, and also perform the counts.
To graph and obtain your statistics you could use graphpad, if you do not have experience in its use, you can look for tutorials on youtube. Finally, to determine if any variable changes the viability or number of cells in your spheres, you can use trypan blue, MTT, resazurin, MTS counts.
