I am extracting RNA from Blood (50ul) using promega Reliaprep RNA cell miniprep system I measure the absorbance at 260 and got 344ug/ml however when I run the sample on gel, not bands appear or very faint ones. is it logical to have good absorbance but no bands?
Since absorbance seems to indicate a relatively high RNA concentration - it may be also good to think of: i) using a fluorescent dye (e.g. Qubit fluorometer) to measure RNA conc. - in case there was a pollution in the sample or ii) run gel again - making sure, that both agarose and electrophoresis chamber are RNase free. RNA degradation during electrophoresis may be a probable cause of your problem.
Since you extracted total RNA ( correct?) you should see clearly two predominant ribosomal RNA bands and a smear, possibly a high molecular weight band (DNA) if you load about 5 ug of RNA (ie. 10-15 microlites of your RNA). It is important that you do not have degradation during electrophoresis :you need to create RNAase free conditions ( everything RNAase free: loading dye, combs, trays, tank, etc....)
