Hello,

I'm using a Rabbit Glucagon antibody from Abcam to stain human primary islets and iPSC derived islets for Glucagon along with other pancreatic cell markers such as PdX1 and C-peptide. The stain works really well with primary islets but with iPSC derived ones, PdX and C-peptide show up well, but in most of the places glucagon appears to have stained nuclei as well. This is not supposed to happen as both C-peptide and Glucagon are released in the cytosol.

Our current hypothesis to explain this is that in our iPSCs derived islets, the beta cells have matured completely (and show C-peptide) whereas the alpha cells have not, which is why they show up in the nuclei.

Has anybody had similar results? Any suggestions are most welcome.
