Hi all, I am working with Leishmania (V) panamensis and in our transcriptomic analysis we saw that IFN is upregulated in an specific group of parasite strains. Our next step is to set up a functional assay to measure IFN in human macrophages derived from monocites of healthy donors. I was wondering if anyone has measure IFN in functional assays (not ELISA) in these cells and could share with me how was done.

Hi!
From monocytes culture, you may take supernadant to Elisa and cellsto do Real Time PCR, with Taqman assay for TNF.
With this assays, you will see upregulation on expression and the activity upregulated.
With the cells, you may do a Western. Blott to TNF too.
