I am doing an experiment where I induce C8-B4 microglial cells with 10 micrograms of LPS at different volumes. The goal is for these cells to be activated and release glutamate. Can I freeze the conditioned media created from this and will this have any effects on the glutamate? I will eventually use a glutamate assay kit to measure the glutamate levels using spectrophotometry
First of all why freeze? Cannot you measure Glu right after the assay?
Or keep if at 4 degrees for some hours and then measure. Probably not much problem on freezing if completely necessary, but you will have to compare by yourself the same medium before and after freezing.
It is OK to freeze at -80 degree C and, when needed to thaw, a quick thaw at 37 degree C.
Thank you for answering! I'm currently waiting for the microcuvettes that my lab ordered and I didn't want to waste time while waiting, so I started doing the experiments and decided to store them until they arrive.
