I am going to do a nested PCR. and also test with a carrier DNA (salmon sperm DNA stock is 1mg/ml). The protocol suggests using 500ng in the reaction mixture. Do I make a diluted stock from the 1mg/ml? or how do I go about this to make it to a 500ng in my reaction mixture (total volume 20ul)?

By including 0.5µL of your carrier DNA stock in your 20µL reaction mix recipe ! And compare with a control without (ie. with 0.5µL of water instead DNA carrier).

If you make a diluted stock of 10 µg/ml , you can add 1 µl of it to a total PCR reaction volume of 20 µl to get your required concentration of 500 ng/ml.

your stock is 1 mg/ml = 1 ug/ul = 1000 ng/ul. This means 1 ul contains 1000 ng.
So to add 500 ng you need 0.5 ul of your stock.
