Not really. We have used them once annealed and aliquoted. An easy way to anneal them is to use a PCR machine and tell it to ramp down from 65C at 1C per min to 4C or 25C. Take about 60 mins. In fact, if it worries you to much you can set up the PCR machine to anneal while you prepare the rest of the expt. If I have not used them in a long time, I juts reanneal the aliquot.
Step wise cooling is better than just keeping at RT.
