Preparation sample for SAM, samples were fixed with glutaraldehyde and then post-fixed with OsO4. OsO4 is extremely toxic to human and damage proteins and other components of sample. Could I skip this step in SEM preparation sample? My samples are rice leaf of 7-day-old seedling.

It is not clear what do you want to see amd which methods in SEM you want to use to observe your sample.
If it is only topography you dont need to stain it with OsO4.
If you want to do some 3D reconstruction and you you need to stain it with OsO4 or get some idea from this webpage (enclosed).
But all the chamicals are toxic in osme way I suppose

Dear Tuyet.
As you know, these dyes will enhance the BSE signal from the membranes (phospholipids). So if you want to see the structures in crossection and want to have the membrane contrast enhanced you need dyes.
But if you want to observe the outermost surface of the leaf, you dont need to enhance anything, but you need to prepare the sample for SEM in order to prevent drying of the sample or use the ESEM - (water vapours in SEM).
